Activated Prothrombin Time (APTT):
Activated prothrombin time test is a type of blood test used to measure the time taken for the citrated plasma to clot in the presence of a surface activator, calcium chloride, and phospholipid.
Materials required:
- Test Tube (Vaccutainer)
- Patient's Plasma (Obtainted by centrifugation of blood sampe)
- Anticoagulant
- Surface Activator
- Phospholipid
- Calcium chloride
- Sterile Glass Stirrer
- Optical Or Mechanical Instrument
- Stopwatch.
- Collect the patient's blood sample in a clean vacctainer containing anticoagulant.
- Centriguge the blood sample under certain rpm in given time 3-5 minutes.
- Then separate the supernatant plasma from the whole blood in a separate test tube.
- Take 0.1 ml of plasma in a sterile test tube and add 0.1 ml of surface activator kaolin to it and mix it throughly.
- Further, the mixture should be placed in a water bath for 10 minutes at 37 degree centigrade.
- Then in other test tube mix 2 ml of calcium chloride solution and 2 ml phospholipids suspension and incubate in a water bath for 10 minutes at 37 degree centigrade.
- Take 0.2 ml of mixture of phospholipid and calcium chloride into the test tube containing mixture of plasma and surface activator kaolin,
- Stir the mixture thoroughly and then immediately on the stop watch.
- Then note the time interval for the mixture to clot.
Normal range of prothrombin test: 30-40 seconds.
Uses Of Prothrombin Test:
- It is used to monitor the heparin therapy.
- It is used as screening test to detect bleeding disorders due to deficiency of the coagulation factors such as factor II, VIII, IX, X, XI, and XII.
- It helps the physician to diagnose and direct the treatment for the disorder.
